## Executive GIST (TL;DR)
- This preprint reports a comparative analysis of the **gut microbiome** in 157 Vietnamese women: 76 urban and 81 rural residents, using shotgun metagenomic sequencing. - The study evaluated alpha and beta diversity, taxonomic composition, and metabolic pathway abundance between rural and urban groups. - In age‑adjusted models, rural participants had higher **alpha diversity** and different **beta diversity** (p 0.1), indicating those factors may mediate observed differences. - In the fully adjusted model, 17 microbial taxa (including Ruminococcus sp AF13_28 and Prevotella pectinovora) and three metabolic pathways remained significantly different (FDR < 0.1). - The authors conclude that **urbanization‑related factors** likely contribute to gut microbiome differences between urban and rural Vietnamese women. - The study was approved by IRBs at the Vietnam National Cancer Institute and Vanderbilt University Medical Center; data are available on request from the corresponding author. - Funding sources included the National Cancer Institute (P20CA210300) and OISE-19-66185-1. The manuscript is a preprint and has not undergone peer review.

Urbanization has been suggested to alter the gut microbiome. We comprehensively evaluated differences in gut microbiome profiles of 76 urban and 81 rural female residents of Vietnam using shotgun metagenomic data. We evaluated rural-urban differences in gut microbial diversity, individual microbial taxa, and metabolic pathways. Rural residents had higher alpha and beta diversities in age-adjusted models (p 0.1). In the fully adjusted model, significant rural-urban differences were observed in the abundance of 17 microbial taxa, including Ruminococcus sp AF13 28 and Prevotella pectinovora, as well as in three metabolic pathways (FDR<0.1). This study suggests that urbanization-related factors may contribute to differences in the gut microbiome between urban and rural Vietnamese women.
