## Executive GIST (TL;DR)
- The study reports that the Arabidopsis pollen-specific protein **ANN5** localizes to the **nucleolus** in pollen and exerts dose-dependent effects on ribosome biogenesis. - Investigators observed altered steady-state **rRNA** levels in mature pollen when ANN5 levels were changed, indicating a regulatory role in rRNA accumulation. - The study team expressed ANN5 ectopically in somatic tissues and recorded accumulation of translationally repressed RNA species in those tissues. - Biochemical data indicated physical interactions between **ANN5** and two epigenetic modulators: histone deacetylase **HD2B** and **NUC1** (nucleolin), both linked to ribosome biogenesis control. - In metabolically active cells, ANN5 associated with ribosomal and ribosome-associated proteins, suggesting a role in modulating translation-related protein networks. - Authors propose ANN5 coordinates formation and availability of pollen RNA reserves during maturation and activation, functioning as a regulator of the pollen “sequestrome.” - The report did not provide detailed descriptions of some experimental parameters (for example, exact assays, quantitative methods, or statistical measures are not fully enumerated in the source abstract). - Funding was declared (National Science Center, Poland OPUS 22); the authors declared no competing interests. - The preprint is indexed on bioRxiv (doi provided) and represents new-results reporting; peer review status beyond posting was not stated in the source.

The molecular mechanisms governing the accumulation of these RNA reservoirs remain poorly understood. Here, we show that the Arabidopsis pollen-specific protein annexin 5 (ANN5) localizes to the nucleolus and exerts dose-dependent control over ribosome biogenesis, as evidenced by altered steady-state rRNA levels in mature pollen. Ectopic expression of ANN5 also induced the accumulation of translationally repressed RNA in plant somatic tissues. Mechanistically, ANN5 interacts with histone deacetylase 2B (HD2B) and nucleolin (NUC1), which are known epigenetic modulators of ribosome biogenesis. In metabolically active cells, ANN5 associates with ribosomal and ribosome-associated proteins, suggesting it modulates the regulatory landscape of translation. We propose that the primary physiological role of ANN5 is to coordinate the formation and subsequent availability of RNA reserves during pollen maturation and activation.
