## Executive GIST (TL;DR)
- The study reports a genetically programmable hydrogel platform assembled with **isopeptide chemistry** that enables molecularly defined extracellular matrix (ECM) mimics. - Investigators engineered modular protein crosslinkers that include complete folded ECM domains and terminal **SpyTag** peptides; those crosslinkers were integrated into **SpyCatcher**-functionalized natural polysaccharide backbones. - The platform separates and independently controls matrix composition, bulk mechanics, viscoelasticity, and biochemical signaling through interchangeable components. - Point mutations introduced into crosslinker SpyTags altered the bond interaction within the SpyTag–SpyCatcher interface, shifting behavior from covalent-like to more physical bonding; the authors reported this changed stress-relaxation without changing matrix composition or stiffness. - Using colorectal cancer models, the team reported that both polymer identity and matrix viscoelasticity independently influenced spheroid growth, invasion potential, and cell dispersion. - Recombinant human ECM domains retained native adhesion functions in the assembled hydrogels, providing a structurally encoded layer of biological information distinct from matrix mechanics. - The work was described in a preprint; the authors have filed a related PCT patent application and the manuscript is not peer reviewed. - The source did not report detailed numerical results, experimental sample sizes, or specific methods for mechanical testing and cell assays within the abstract; those details appear in supplementary material or full text and are not reproduced here.

Here, we present a genetically programmable hydrogel platform that enables independent control of mechanical and biological properties including matrix composition, mechanics, viscoelasticity, and biochemical signaling using recombinant human ECM building blocks assembled via isopeptide chemistry. Modular protein ECM crosslinkers containing complete folded ECM domains equipped with terminal SpyTag peptides were integrated within SpyCatcher-functionalized interchangeable natural polysaccharide backbones to generate fully defined matrices with decoupled material parameters. Point mutations on crosslinker SpyTags within the bond interface varied bond affinity from covalent to physical bonding, allowing tuned stress-relaxation behaviors across physiologically relevant regimes without altering matrix composition nor stiffness. Using colorectal cancer models, we show that polymer identity and matrix viscoelasticity independently regulate spheroid growth, invasion capabilities, and dispersion. Importantly, recombinant ECM domains recapitulated native adhesion functions and provide a structurally encoded layer of biological information. This hydrogel platform provides a molecular framework for constructing precision recombinant human ECM mimics with broad applicability across biomaterials science, mechanobiology, disease modeling, tissue engineering, and regenerative medicine.
