## Executive GIST (TL;DR)
- Investigators surveyed wild-type strains of **D. mojavensis** and related species and found unexpectedly high rates of sex-chromosome aneuploidy, with some strains carrying **XXY** and **XXYY** females at frequencies up to 88%. - Historical context: Bridges reported rare fertile **XXY** females in D. melanogaster (~1/2000), but the current cohort shows far higher occurrence in these species. - The cohort included males frequently observed as **XYY** or XYYY in the same strains; wild laboratory strains showed these karyotypic compositions at variable frequencies. - Field sampling was limited by population declines and safety concerns, but investigators detected one **XXY** female (1/87) and one **XYY** male (1/36) in a natural population, demonstrating that these genotypes appear in nature as well. - Eggs from **XXY** females were expected to be 50:50 XY:X-bearing; empirical data showed a 57:43 ratio, indicating a departure from Mendelian expectation. - The study team applied mathematical modeling and reported that the observed meiotic-drive advantage (57:43) is sufficient to account for the elevated frequencies of **XXY** females in laboratory strains. - Authors argued that Y-chromosome evolution in these taxa may be influenced by female-phase fitness when **XXY** females are common, since the Y contributes to female genotypes in these cases. - The source reported limitations: constrained field collections and safety issues; specific experimental methods and sampling protocols were not detailed in the provided text.

Although widely used in genetic studies, XXY females are seldom found in wild-type strains or natural populations, presumably because natural selection keeps them at very low frequency. Here we show that many wild-type strains of D. mojavensis and its close relatives D. arizonae, D. navojoa, and D. wheeleri carry XXY and XXYY females at frequencies of up to 88%; males from these strains are frequently XYY or XYYY. Although field collections were hampered by population declines and safety concerns, we found one XXY female (out of 87) and one XYY male (out of 36) in a natural population of D. mojavensis, indicating that these genotypes also occur at appreciable frequencies in nature. XXY females are expected to produce XY-bearing and X-bearing eggs in equal proportions; instead, we found a 57:43 ratio. Mathematical modeling shows that this meiotic-drive advantage is sufficient to explain high frequencies of XXY females. It is universally accepted that Y evolution responds exclusively to male selection, yet in XXY-bearing species it also responds to female fitness. ### Competing Interest Statement The authors have declared no competing interest. ## Funder Information Declared Wellcome Trust, https://ror.org/029chgv08, 207486Z17Z Copyright The copyright holder for this preprint is the author/funder, who has granted bioRxiv a license to display the preprint in perpetuity.
