## Executive GIST (TL;DR)
- The authors present a **rapid, reliable, and highly reproducible soil-based screening protocol** to evaluate drought tolerance at the seedling stage across diverse small grain cereals and their wild relatives. - The protocol is designed for high-throughput evaluation and was used to screen up to **1200 genotypes** spanning wheat, oat, barley, and wild relatives including **Aegilops tauschii** and **Ae. umbellulata**. - The method effectively distinguishes **drought-tolerant** versus **drought-sensitive** genotypes while minimizing environmental variability in the screening environment. - Two contrasting wheat genotypes identified by this seedling-stage screen were further evaluated under drought imposed at **anthesis and grain filling**, and their contrasting responses were maintained at later developmental stages. - The protocol is proposed as a practical preliminary screening tool to identify contrasting genotypes for downstream **breeding**, physiological, and genetic studies. - The study is reported as a preprint (bioRxiv) and funded by the USDA Agricultural Research Service; it has not undergone peer review. - Specific experimental details (exact soil composition, drought regime parameters, scoring metrics, growth conditions, and statistical analyses) are not described in the source summary and therefore are not reported here.

Improving genetic tolerance to drought stress requires detailed characterization of global germplasm collections and their wild relatives under drought conditions. However, the lack of rapid screening methods remains a major obstacle in achieving this goal. Here, we present a rapid, reliable, and highly reproducible soil-based screening protocol for evaluating large germplasm collections under drought stress at the seedling stage. This protocol efficiently discriminates drought tolerant and sensitive genotypes with minimal environmental variability and is broadly applicable across various small grain cereals, enabling the evaluation of 1200 genotypes of wheat, oat, barley, and their wild relatives such as Aegilops tauschii and Ae. umbellulata. Additionally, evaluation of two contrasting wheat genotypes identified using this protocol under drought stress imposed at anthesis and grain filling stages provided evidence that their contrasting drought responses were also consistently maintained at the later developmental stages. Given its efficiency, reproducibility, and cross-species applicability, this protocol offers a practical approach for preliminary screening of large germplasm collections as well as for identifying contrasting genotypes to be included in subsequent breeding, physiological, and genetic studies. ### Competing Interest Statement The authors have declared no competing interest. ## Funder Information Declared USDA Agricultural Research Service Copyright The copyright holder for this preprint is the author/funder, who has granted bioRxiv a license to display the preprint in perpetuity.
