## Executive GIST (TL;DR)
- The review article examined how **cytokine** networks coordinate interactions among neurons, glia, resident macrophages (microglia and border-associated macrophages) and infiltrating leukocytes to maintain CNS integrity. - Authors reported that these circuits set a basal immune tone, influence responses to perturbation and drive tissue adaptation and recovery across diverse CNS states. - The review focused on specific homeostatic cytokines: **CSF-1**, **IL-34**, **TGFβ** and **IL-33**, noting their roles in microglial development, maintenance and quiescence according to the figures and text cited. - Investigators outlined inflammatory mediators — **IL-1**, **TNF**, **GM-CSF** and **IFNγ** — as drivers of CNS inflammation and as modulators of immune cell function during tissue damage. - The study team examined type I and type II **interferons** for dual roles in antiviral defense and in neurodegenerative states, referencing pleiotropic regulatory effects depicted in the figures. - The authors positioned **IL-12** at the interface of innate sensing, adaptive immunity and tissue-protective adaptation, proposing it as an instructive module within cytokine networks. - The review used illustrative figures: CSF-1R/TGFβ signaling for microglia (Fig. 1); IFN-I/IFNγ roles (Fig. 2); GM-CSF and neuronal IL-12 signaling effects (Fig. 3). - Data indicated that cytokine miscalibration can contribute to neurological disease across contexts; the review synthesized organizing principles rather than presenting new experimental datasets. - The article is a Review published in Nature Immunology (2026); the source provided an abstract and figure legends but access to full subscription content was noted by the publisher. - The review highlighted instructive modules to frame how cytokine networks preserve, destabilize and restore CNS integrity, and it cited foundational and recent primary research across the field.

These circuits establish basal immune tone, shape responses to perturbation, and promote tissue adaptation and recovery. In this Review, we examine selected cytokine pathways across homeostasis, inflammatory or degenerative tissue damage and repair to develop a framework for how cytokine networks preserve, destabilize and restore CNS integrity. We focus on CSF-1, IL-34, TGFβ and IL-33 in homeostasis; IL-1, TNF, GM-CSF and IFNγ in inflammation; type I and II interferons in antiviral and neurodegenerative states; and IL-12 at the interface of innate sensing, adaptive immunity and tissue-protective adaptation. By focusing on instructive modules, we highlight broader organizing principles and how cytokine miscalibration can lead to neurological disease across diverse contexts.
