## Executive GIST (TL;DR)
- The authors re-analyzed RNA-seq from outer hair cells (OHCs) to test whether **honokiol** effects on OHC gene expression differ with **cisplatin** exposure. - Twenty-two genes tied to mechanotransduction, hair bundle function, ion homeostasis, electromotility, and efferent regulation were selected a priori for focused analysis. - Interaction statistics were available for 20 of those genes; 16 showed significant Honokiol × Cisplatin interaction effects, and 15 of those interactions were negative. - In the context of **cisplatin**, honokiol downregulated several mechanotransduction (MET) machinery genes, specifically Lhfpl5, Tmc1, Cib2, and Tomt, according to the re-analysis. - The authors note a potential mechanistic link: MET channels have been implicated in cisplatin entry into hair cells, so altered MET gene expression might contribute to honokiol-mediated protection against cisplatin-induced hearing loss. - The dataset supporting these findings is available at GEO accession GSE328003. - The study was funded in part by the National Institute on Deafness and Other Communication Disorders (R01 DC019434). - The authors explicitly state that whether the observed transcriptional changes reduce cisplatin uptake was not determined in the reported analysis. - No competing interests were declared by the authors. - The report is a preprint posted on bioRxiv; it is not indicated whether additional functional assays, dosing, or temporal details were reported beyond the RNA-seq re-analysis.

We previously reported that honokiol protects against CIHL and mitigates cisplatin-induced transcriptomic changes in OHCs. Here, we re-analyzed OHC RNA-seq to determine whether honokiol differentially affects genes involved in OHC physiological function depending on cisplatin exposure. Twenty-two genes involved in mechanotransduction and hair bundle function, membrane potential and ion homeostasis, electromotility, and efferent regulation were selected. Among 20 genes with available interaction statistics, 16 showed significant Honokiol x Cisplatin interactions, of which 15 were negative. Notably, in the presence of cisplatin, honokiol downregulated several genes associated with mechanotransduction (MET) machinery, including Lhfpl5, Tmc1, Cib2, and Tomt. Because MET channels have been implicated in cisplatin entry into hair cells, these changes raise the possibility that altered MET machinery may contribute to honokiol-mediated protection against CIHL. However, whether these changes decrease cisplatin uptake remains to be determined. Competing Interest Statement The authors have declared no competing interest.
