## Executive GIST (TL;DR)
- The study evaluated genetic associations with resting-state **EEG** features in 169 Finnish participants spanning subjective cognitive decline to mild cognitive impairment (MCI). - EEG metrics included periodic **alpha- and beta-band** features and aperiodic spectral parameters derived from eyes-closed recordings. - Genome-wide genotypes were QC'd, imputed and functionally annotated, yielding 16,935 gene regions and 90,607 functional variants for analysis. - Associations between genetic variation and latent EEG features were tested using Bayesian reduced rank regression (BRRR). - At P < 0.005, 145 genes were associated with **periodic** EEG features and 39 genes with **aperiodic** EEG features; however, no associations survived correction for multiple testing. - Candidate genes grouped around pathways involved in **synaptic transmission**, neuronal excitability and neurodevelopment; examples include RASGEF1C and TREML2 at loci previously linked to Alzheimer’s disease (AD). - Aperiodic-feature candidate genes showed greater enrichment for **neuroinflammatory** processes (e.g., C5AR2 within a FinnGen AD-associated region) compared with periodic-feature candidates, which were more often related to intracellular neuronal maintenance and signaling. - Unsupervised clustering based on periodic EEG–associated variants produced participant subgroups that differed significantly in plasma **p-tau217** concentrations and delayed verbal recall after multiple-testing correction. - Top genes driving cluster separation included **ACAN, INPP5B, CAMKK2, CABIN1,** and **SPATA13**. - The authors interpret the results as indicating that periodic and aperiodic EEG features capture partly distinct biological processes within the MCI continuum and that EEG-derived endophenotypes can help dissect heterogeneity in early cognitive decline and AD-related pathology. - Limitations reported in the source: reported gene associations did not survive multiple-testing correction; raw brain imaging data cannot be shared openly due to ethics restrictions, although derived models and code are available at the BioMag GitLab repository. - Ethical approvals and participant consent were obtained from the HUS Regional Medical Research Ethics Committee, Helsinki University Hospital. Funding sources included several Finnish foundations and EU programmes as listed in the source.

Electroencephalography (EEG)-derived features as heritable markers of neuronal network activity may provide biologically informative endophenotypes for studying cognitive decline in the MCI. We investigated associations between functional genetic variation and resting-state EEG features in 169 Finnish individuals with symptoms spanning from subjective cognitive decline to MCI. Periodic alpha- and beta-band features and aperiodic spectral parameters were derived from eyes-closed EEG recordings. Genome-wide genotypes were quality-controlled, imputed, functionally annotated (yielding 16,935 gene regions comprising 90,607 functional variants), and associated with latent EEG features using Bayesian reduced rank regression. We integrated the results with neurobiology-related literature review and known AD-associated loci and performed unsupervised clustering of participants based on EEG-associated genetic variation, testing clinical and biomarker differences between clusters. We identified 145 genes associated with periodic EEG features and 39 genes with aperiodic EEG features at P ## Funder Information Declared Emil Aaltonen Foundation Institutional research funding from Helsinki University Hospital (HUS), Y780023059, Y780023086, Y780024065, Y780024067, Y780025056, Y780026002 Sigrid Jusélius Foundation, https://ror.org/00ckakm23 Academy of Finland, grant numbers 321460 and 355409 and the Flagship of Advanced Mathematics for Sensing Imaging and Modelling grant 359181 Finnish Cultural Foundation Finnish Research Impact Foundation Finnish Ministry of Education and Culture’s Doctoral Pilot for Mathematics in Sensing, Imaging and Modelling Instrumentarium Science Foundation European Union (Horizon Europe programme), 101155955 EU Horizon 2020 research and innovation pr
