- The study screened genomes from 263 Latin American individuals with **early-onset Parkinson's disease (EOPD)** and/or a positive family history who lacked known pathogenic PD variants on prior genotyping. - Authors performed short-read whole-genome sequencing to examine seven primary PD genes and 21 genes linked to related parkinsonian disorders. - Background context: PD is typically age-related and sporadic, but **5–14%** of people have onset before age 50 and about **20%** report family history, both groups with increased likelihood of genetic contribution. - Findings: a **monogenic burden** (pathogenic or likely pathogenic variants in primary and secondary PD genes) was observed in **4.7%** of sequenced individuals. - Additional results: **5.5%** carried pathogenic or likely pathogenic variants for **Gaucher's disease**, and **2.7%** had known risk variants in **GBA1**. - The study expanded the catalogue of disease-associated variants in a Latin American cohort, identifying variants that may be missed by array-based or targeted assays. - Data access: study data are available from the corresponding author on request and will be released through the Global Parkinson's Genetics Program (GP2) in a future release. - Ethical oversight: Cleveland Clinic Institutional Review Board approval (IRB #19-340) and participant consent were obtained. - Funding sources included NIH grants, Michael J.
