## Executive GIST (TL;DR)
- The study generated a fully humanized **ATN1** mouse carrying a pathogenic CAG expansion (112 repeats) to model dentatorubral‑pallidoluysian atrophy (**DRPLA**) seizure phenotypes. - Investigators compared Atn1Q112/+ mice to wild‑type controls after neonatal (P2) and 4‑week interventions with an investigational antisense oligonucleotide (**ASO**) targeting human, CAG‑expanded **ATN1**, or saline. - The cohort underwent acute seizure testing, ataxia profiling, and chronic video‑EEG monitoring for spontaneous seizures through 8 weeks of age. - Saline‑treated Atn1Q112/+ mice showed increased seizure susceptibility and earlier mortality relative to wild‑type mice. - ASO treatment in Atn1Q112/+ mice normalized seizure threshold in acute testing, reduced ataxia measures, and lowered early death risk compared with saline‑treated mutants. - Chronic video‑EEG detected spontaneous behavioral seizures in untreated Atn1Q112/+ mice across two weeks of recording; these events were absent in ASO‑treated Atn1Q112/+ mice and in wild‑type animals. - The investigators observed a mutation‑driven decrease in delta and theta EEG frequencies in Atn1Q112/+ mice; ASO treatment prevented the recorded spontaneous seizures. - The report presents Atn1Q112/+ mice as a highly seizure‑prone DRPLA model, and posits **ATN1** as an epilepsy‑related risk factor and a therapeutic target for antisense strategies. - The preprint did not report specific ASO dose amounts or detailed safety pharmacology; authors declared no competing interests and noted funding sources including CureDRPLA and the American Epilepsy Society.

Mutant ATN1 (mATN1) expression results in symptoms that inversely correlate to CAG-expansion size: young-onset cases have the most CAG repeats and developmental delay, high-frequency treatment-resistant seizures, and early mortality. We sought to characterize the seizure susceptibility of a fully humanized ATN1 mouse expressing human mATN1 with 112 CAG repeats to then assess therapeutic antisense oligonucleotide (ASO) intervention on seizure-related outcomes. Atn1Q112/+ or wild-type (WT) mice were treated with investigational ASO targeting human, CAG-expanded, ATN1 or saline (P2 and then again at 4 weeks-old) before acute seizure testing, ataxia profiling, and chronic in vivo video-EEG recording for spontaneous seizures up to 8 weeks-old. Saline-treated Atn1Q112/+ mice were more seizure prone with early death. However, ASO administration to Atn1Q112/+ mice normalized seizure threshold, decreased ataxia, and protected against early death. Atn1Q112/+ mice exhibited spontaneous behavioral seizures recorded over 2 weeks, which were absent in ASO-treated Atn1Q112/+ and WT mice, with a specific mATN1 expression-driven decrease in delta and theta EEG frequencies. Atn1Q112/+ mice are highly seizure prone, validating this model for DRPLA epilepsy. ATN1 represents a novel epilepsy-related risk factor for therapeutic investigation. ### Competing Interest Statement The authors have declared no competing interest. ## Funder Information Declared CureDRPLA American Epilepsy Society, https://ror.org/01sx4nq39, N/A - 2024 Seed Grant Silva Ataxia Foundation University of Washington, Department of Pharmaceutics Copyright The copyright holder for this preprint is the author/funder, who has granted bioRxiv a license to display the preprint in perpetuity.
