- Background: Autism spectrum disorder features prominent social difficulties. Preclinical rodent work implicates oxytocin in modulating social behaviors via a distributed **social salience network** (SSN), but whether a homologous SSN exists and is clinically relevant in humans had not been established. - Objective: The authors retrospectively examined a resting-state fMRI dataset from a prior randomized, placebo-controlled intranasal **oxytocin** trial to test whether a human SSN can be defined, whether SSN functional connectivity (FC) relates to social and repetitive behaviors, and whether intranasal oxytocin modulates SSN connectivity in adult men with **autism**. - Methods: A human SSN atlas was assembled from open-access cortical and subcortical parcellations and used to compute ROI-to-ROI resting-state FC across the network. Analyses assessed SSN-wide and sub-network FC correlations with behavioral trait measures and with salivary oxytocin concentrations at baseline and after administration; treatment effects were tested using ANCOVA. - Key baseline findings: Specific SSN sub-network FC correlated with measures of social and repetitive behaviors. Circuits linking somatosensory cortex with the **nucleus accumbens** and with the **paraventricular nucleus** showed marked sensitivity to oxytocin at baseline (salivary concentrations linked to FC). - Treatment-induced changes: After intranasal oxytocin administration, the most prominent FC increase was observed in an **amygdala–somatosensory** circuit. This strengthening represented the largest oxytocin-induced shift within the SSN. - Predictive biomarker potential: Baseline connectivity within the amygdala–somatosensory circuit strongly predicted the magnitude of post-treatment FC change: individuals with lower baseline FC exhibited larger increases after oxytocin. - Clinical implication: The study provides first evidence supporting the relevance of a human SSN in autism and identifies specific circuits that may serve as candidate biomarkers to predict responsiveness to intranasal oxytocin. - Limitations and transparency: The analysis was retrospective, based on a previously published randomized, placebo-controlled trial; this preprint has not been peer reviewed. Oxytocin supplementation shows promising therapeutic efficacy in alleviating autism-like traits in rodent models, but clinical effects in humans remain inconsistent. The rodent-derived social salience network (SSN) comprises several oxytocin-modulated brain regions implicated in social behavior, but its conservation has not been established in humans. Here we assess, for the first time, functional connectivity (FC) within a homologous human SSN in autistic men to examine its relationship with behavioral traits and modulation by oxytocin. Methods: The human SSN atlas was collated from open-access cortical and subcortical parcellations, and used to retrospectively analyze a resting-state fMRI dataset of adult men with autism from a previously published, randomized, placebo-controlled oxytocin trial. SSN-wide and sub-network ROI-to-ROI FC correlations with social trait expression and salivary oxytocin concentrations were performed at baseline and post-administration. Treatment specific outcomes on FC were calculated using ANCOVA. Results: We observed SSN sub-network FC correlations with social and repetitive behavioral scores and identified strong oxytocin sensitivity of nucleus accumbens-somatosensory and paraventricular nucleus-somatosensory circuits at baseline. Following nasal spray administration, a strengthening of amygdala-somatosensory circuit was detected as the largest oxytocin-induced FC shift. Notably, baseline connectivity within this circuit strongly predicted treatment response, with individuals having lower baseline FC showing greater post-treatment FC.
