## Executive GIST (TL;DR)
- The authors conducted a coordinate-based meta-analysis of voxel-based morphometry (VBM) studies to identify common and disorder-specific gray matter volume (GMV) deficits across **schizophrenia**, **major depressive disorder**, and **alcohol use disorder**. - The literature search covered PubMed, Web of Science and PsycInfo from January 2000 to March 2024, with adherence to PRISMA and a pre-registered analysis plan on OSF. - A total of 147 VBM studies were included, representing 90 schizophrenia, 44 MDD and 13 AUD studies; pooled sample size was reported as 13,041 participants. - Seed-based d Mapping with Permutation of Subject Images (SDM-PSI) and a random-effects model generated statistical brain maps; inference used permutation testing with p < .0025 and cluster extent ≥10 voxels. - Shared GMV reductions were identified in the **ventromedial prefrontal cortex**, ventral anterior cingulate cortex, insula and superior temporal gyrus across the three disorders. - Network-level analyses linked these common regions to cortical midline structures and social cognition / theory-of-mind behavioral functions, indicating functional interactions beyond local atrophy. - Additional characterization implicated dopaminergic and serotonergic neurotransmitter systems and related genetic annotations in the shared regions. - Heterogeneity across studies was evaluated with the I2 statistic; publication bias was tested using Egger's regression — the source reports these evaluations but does not detail specific I2 values or publication-bias outcomes in the abstract. - The sample composition was imbalanced across disorders (many more SZ studies than AUD), a factual feature of the included literature; the source did not report further subgroup or dosing details in the abstract. - Data and analytic materials are available on OSF at the cited pre-registration link; authors declared no competing interests.

Objective: To determine common and disorder-specific gray matter deficits in patients with schizophrenia (SZ), major depression disorder (MDD) and alcohol use disorder (AUD) compared to heathy controls. Data Sources: A comprehensive literature search using three databases including PubMed, Web of Science and PsycInfo (between January 2000 to March 2024) as well as searches on review articles. Study Selection: Studies had to be published in English, reporting whole-brain results, comparing patients and healthy controls. Data Extraction and Synthesis: This meta-analysis adhered to the PRISMA guidelines and was pre-registered (https://osf.io/s57uq) prior to data analysis. Data was independently extracted and verified by 3 contributors, and statistical brain maps were generated using Seed-based d Mapping with Permutation of Subject Images (SDM-PSI) random-effects model. 147 VBM studies encompassing 90 Schizophrenia, 44 MDD and 13 AUD studies were finally included. Main Outcomes and Measures: Statistical maps of common and disorder-specific gray matter deficits in patients versus healthy controls are presented. Statistical inference utilized permutation tests, with threshold at p The Chan Zuckerberg Initiative, Cold Spring Harbor Laboratory, the Sergey Brin Family Foundation, California Institute of Technology, Centre National de la Recherche Scientifique, Fred Hutchinson Cancer Center, Imperial College London, Massachusetts Institute of Technology, Stanford University, The University of Edinburgh, University of Washington, and Vrije Universiteit Amsterdam.
