## Executive GIST (TL;DR)
- Investigators designed length-encoded **micropillar arrays** in which pillar stiffness is determined by effective pillar length while pillar tops remain level and evenly spaced, enabling patterned stiffness that varies at cell scale. - Pillar diameters of 2, 2.5 and 3 µm produced stiffness values spanning 4.0–82.6 nN/µm, reported as an effective modulus range of 2.9–39.4 kPa; variation occurred periodically over distances comparable to single-cell dimensions. - The platform permits simultaneous readouts of cell morphology, alignment, spreading, pillar displacement, and traction forces, as the study team reported. - In experiments with **MDA-MB-231** breast cancer cells, pillar displacement showed a biphasic response: displacement fell with increasing stiffness up to ~10 nN/µm, then stabilized near 0.5 µm at higher stiffness. - Traction forces, by contrast, increased continuously across the full measured stiffness range; the cohort thus separated displacement and force responses across stiffness regimes. - Cells contacted soft pillars more frequently than stiff ones, with approximately 23% greater coverage of soft regions, indicating contact preference tied to local compliance. - Cells avoided large stiffness mismatches along their length: when both cell ends sat on pillars of equal stiffness, 48–50% of cells lay along a single stiffness row versus ~14% expected by chance, according to the authors. - Cell alignment clustered along two principal array axes (27–31% of cells within 10°), and alignment rose modestly with increasing pillar diameter. - The authors argue the arrays offer a tool to study how individual cancer cells sense steep, local mechanical heterogeneity relevant to tumour microenvironments. - The preprint is not peer-reviewed; the abstract does not report sample sizes, full experimental protocols, or statistical details, which the source did not provide in that section.

How cells sense and respond to such steep, local stiffness gradients is not well understood. Here, we developed length-encoded micropillar arrays in which stiffness is set by the effective pillar length, while the pillar tops remain at the same height and spacing. Pillars with diameters of 2, 2.5 and 3 µm gave stiffness values of 4.0–82.6 nN/µm (effective modulus 2.9–39.4 kPa), varying periodically over distances comparable to the size of individual cancer cells. This platform allows simultaneous measurement of cell morphology, alignment, spreading, pillar displacement, and traction forces. In MDA-MB-231 breast cancer cells, pillar displacement showed a biphasic response. It decreased with increasing stiffness up to about 10 nN/µm and then remained close to 0.5 µm. In contrast, traction forces increased across the full stiffness range. Cells contacted soft pillars more than stiff pillars, with about 23% greater coverage of soft regions. Cells also avoided large stiffness differences along their length: among cells whose two ends sat on pillars of equal stiffness, 48–50% lay along a single stiffness row, compared with about 14% expected by chance. Cell alignment was enriched along the two principal array directions (27–31% of cells within 10°) and increased slightly with pillar diameter. Length-encoded micropillar arrays therefore provide a platform for studying how cells respond to cell-scale mechanical heterogeneity, with relevance to cancer invasion. ### Competing Interest Statement The authors have declared no competing interest. ## Funder Information Declared Australian Research Council (ARC), DE220100205 Copyright The copyright holder for this preprint is the author/funder, who has granted bioRxiv a license to display the preprint in perpetuity.
