## Executive GIST (TL;DR)
- Early and sensitive detection of colorectal cancer biomarkers remains clinically important; this study demonstrates a novel visual detection platform for **miRNA-126**. - The authors developed a hydrogel kit in which **Pepper RNA** bearing an acrydite group (acrydite-Pepper) is covalently immobilized into the polymer network by photopolymerization. - The immobilized Pepper RNA acts simultaneously as the **capture probe** and the **signal output probe**, forming a target-driven **fluorogenic RNA switch**. - Binding of target **miRNA-126** induces a conformational change in the RNA switch via Watson-Crick base pairing, producing a fluorescent “light-up” signal without additional labeling. - Reported analytical sensitivity (limit of detection) differs by readout: **23.8 pM** using fluorescence microscopy and **0.53 pM** using a microplate reader. - The platform is presented as simple-to-operate, low-cost, and easily visualized, supporting rapid on-site detection and potential preliminary point-of-care testing (**POCT**) for diluted biological samples. - The work is reported as the first immobilization of acrydite-modified Pepper RNA into a hydrogel for target-triggered visual miRNA detection. - Authors declare no competing financial interests or personal relationships influencing the work.

Herein, a novel visual detection platform based on target-driven fluorogenic RNA switch lighting-up for label free measurement of miRNA-126 is demonstrated. For the first time, Pepper RNA with an acrydite group (acrydite-Pepper) is covalently immobilized into the hydrogel network via photopolymerization. The fluorogenic RNA switch simultaneously serves as both a capture probe and a signal output probe, and the conformational change of the switch is induced in the presence of target miRNA through Watson-Crick base complementary pairing. This Pepper RNA functionalized hydrogel kit exhibits a limit of detection (LOD) of 23.8 pM and 0.53 pM for the different detection platform of fluorescence microscopic and microplate reader, making it suitable for rapid and sensitive on-site detection. This strategy provides a simple-to-operate, low-cost, and easily visualized new method for the detection of colorectal cancer-related miRNA, laying a foundation for future applications in diluted biological sample analysis and preliminary point-of-care testing (POCT). **Keywords:** Colorectal cancer; Fluorescence sensing; Hydrogel; Visualization; miRNA-126.
