## Executive GIST (TL;DR)
- Small‑cell lung cancer (SCLC) retains a poor prognosis despite recent advances with **anti‑PD‑(L)1 antibodies** and the DLL3 T cell‑engager **tarlatamab**, creating a need for new therapies. - The phase III TAISHAN‑302 randomized trial compared **tambotatug pelitecan (Tam‑Peli)**, a **B7‑H3‑targeted antibody–drug conjugate (ADC)** with a topoisomerase I inhibitor payload, versus **topotecan** as second‑line therapy for SCLC. - The trial enrolled 451 patients in China whose disease had progressed during or after first‑line platinum‑based chemotherapy; randomization was 1:1 to Tam‑Peli or topotecan. - Most patients (87.1%) had previously received an anti‑PD‑(L)1 antibody as part of first‑line therapy. - Patients were not selected based on **B7‑H3 expression**; biomarker enrichment was not used. - **Overall survival (OS)** was the primary endpoint of the study. - The Nature Reviews Clinical Oncology highlight reports that TAISHAN‑302 results demonstrate promising outcomes with Tam‑Peli, but the review does not report detailed efficacy or safety data or specific numerical results. - The underlying primary publication is cited: Zhao et al., NEJM (2026), which contains the full trial data referenced by the highlight. - The Nature Reviews piece is a research highlight and therefore provides a concise interpretation rather than full trial details; several specifics (exact OS benefit, progression‑free survival, response rates, and safety profile) were not reported in the highlight. - TAISHAN‑302 findings suggest a potential new second‑line option for SCLC, pending full data review and broader context from the NEJM report and regulatory assessment.

Now, results from the phase III TAISHAN-302 trial demonstrate promising outcomes with tambotatug pelitecan (Tam-Peli), a B7-H3-targeted antibody–drug conjugate with a topoisomerase I inhibitor payload, as a second-line treatment for this disease. In this trial performed in China, 451 patients with SCLC that had progressed during or after first-line platinum-based chemotherapy, combined with an anti-PD-(L)1 antibody in 87.1%, were randomly assigned (1:1) to receive Tam-Peli or topotecan. Notably, patients were not selected on the basis of B7-H3 expression. Overall survival (OS) was the primary end point.
