## Executive GIST (TL;DR)
- Investigators generated a murine lung tumor model carrying a **KrasG12D** mutation and compared three Trp53 gene-dosage contexts: intact (K), single-allele loss (KP+/-), and complete loss (KP) using publicly available KP data. - The cohort showed longer survival for K and KP+/- mice compared with KP mice, mirroring clinical observations associating retained wild-type TP53 with better outcomes. - Single-cell transcriptomic profiling of tumor-laden lungs demonstrated that **Trp53** heterozygosity expanded transcriptional plasticity; this expanded plasticity was absent in K and KP tumors. - Genomic instability—measured by copy-number alterations and related profiles—was higher in KP+/- tumors than in KP tumors according to the reported analyses. - Murine KP+/- transcriptional signatures and copy-number profiles were detected in human **NSCLC** datasets, indicating translational relevance of the mouse findings. - Two distinct KP+/- signatures correlated differentially with patient survival in human datasets: one signature associated with better survival and another with worse survival, as the authors reported. - The authors concluded that **TP53** heterozygosity in the context of **KRAS**-mutant lung cancer defines a distinct genomic and phenotypic landscape that could influence therapeutic strategy development. - The source did not provide detailed dosing, therapeutic interventions, or exact single-cell analytic parameters in the abstract; full methods were not included in this summary. - Authors declared no competing interests; the study was funded by The University of Texas MD Anderson Cancer Center and posted on bioRxiv on October 14, 2026.

The most common event at the TP53 locus is mutation/loss of one allele and retention of a wild-type allele, which confers better survival outcomes than complete TP53 loss. To study the contribution of TP53 heterozygosity, we generated a murine lung tumor model with a KrasG12D mutation and intact (K) or single-allele loss (KP+/-) of Trp53 and compared it with publicly available complete loss (KP) data. K and KP+/- mice lived longer than KP mice, paralleling clinical survival data. Single-cell transcriptomic analysis of tumor-burdened lungs indicated that Trp53 heterozygosity expanded transcriptional plasticity that was not present in K and KP tumors. Genomic instability was also higher in KP+/- mice relative to the KP genotype. Trp53 gene dosage, thus, regulates phenotypic plasticity and evolutionary trajectories of tumor states across K, KP+/-, and KP tumors. Murine KP+/- tumor signatures and copy-number profiles were present in human NSCLC datasets. Two of these signatures correlated with better and worse patient survival outcomes, respectively. TP53 heterozygosity in KRAS-mutant lung cancers presents a distinct genomic landscape that should be taken into account in the development of novel therapeutic strategies. ### Competing Interest Statement The authors have declared no competing interest. ## Funder Information Declared The University of Texas MD Anderson Cancer Center, https://ror.org/04twxam07 Copyright The copyright holder for this preprint is the author/funder, who has granted bioRxiv a license to display the preprint in perpetuity.
