## Executive GIST (TL;DR)
- Investigators reanalyzed publicly available **mass cytometry** data from 36 one-year-old infants whose peanut reactivity was confirmed by oral challenge or classified as sensitized tolerance or non-sensitized controls. - The team clustered 3,560,317 unstimulated peripheral blood mononuclear cells into 19 curated immune cell types, using an unsupervised approach to define cellular compartments. - Sensitized-tolerant infants had higher counts of **HLA-DR+ memory CD4 T cells** (1.97-fold increase; q = 0.033) compared with allergic infants. - Lineage-negative CD38+ cells were also expanded in sensitized-tolerant infants (2.26-fold; q = 0.006). - A trend toward more conventional **dendritic cells** in the sensitized-tolerant group was observed (2.55-fold; q = 0.069), and an earlier report of increased **plasmacytoid dendritic cells** in sensitized-tolerant infants was reproduced by the reanalysis. - Activation markers and measured cytokine levels did not differ across groups in the unstimulated samples, according to the reanalyzed dataset. - The authors identified two technical confounders in the mass cytometry dataset: a staining batch imbalance that included half of the sensitized-tolerant samples and CD20 signal spillover into the TNFα channel that varied between staining runs. - Data indicated that the dendritic-cell expansion persisted when restricting to the bright-staining batch, while the T-cell signal weakened in that subset. - The article is a preprint and has not undergone peer review; the authors emphasized the need for specific controls in mass cytometry studies of food allergy to account for batch effects and channel spillover.

Here we reanalyze mass cytometry data from unstimulated peripheral blood mononuclear cells of 36 one-year-old infants with challenge-confirmed peanut allergy, sensitized tolerance or neither. Clustering all 3,560,317 cells into 19 curated cell types, we find that sensitized-tolerant infants have more HLA-DR+ memory CD4 T cells (1.97-fold, q = 0.033) and lineage-negative CD38+ cells (2.26-fold, q = 0.006) than allergic infants, and a trend toward more conventional dendritic cells (2.55-fold, q = 0.069). The dendritic-cell difference persists in samples from the bright-staining batch, whereas the T-cell difference weakens, and the reported expansion of plasmacytoid dendritic cells in sensitized-tolerant infants is reproduced. Activation markers and cytokines do not differ between groups. We also identify two technical confounders: a staining batch that includes half of the sensitized-tolerant samples, and spillover of CD20 signal into the TNFα channel that differs between staining runs. These results link sensitized tolerance to an expanded dendritic-cell compartment and define controls needed in mass cytometry studies of food allergy. ### Competing Interest Statement KIA is the founding CEO of Cytowhiz; the platform on which the publically available data was reanalyzed. ## Footnotes * Copyright The copyright holder for this preprint is the author/funder, who has granted bioRxiv a license to display the preprint in perpetuity.
