## Executive GIST (TL;DR)
- The article is a 2026 review that examined how **acupuncture** modulates multiple signaling pathways implicated in **obesity**, synthesizing preclinical and clinical evidence. - Authors reported that dysregulated glucose and lipid metabolism, insulin and leptin resistance, chronic low-grade inflammation, oxidative stress, and fat accumulation drive obesity pathogenesis. - The review focused on four signaling areas: **AMPK** (energy sensing and fatty acid oxidation), **JAK2/STAT3** (leptin and appetite regulation), **TLR4/NF-κB** (inflammatory signaling), and the **SIRT1** network (metabolic stress and oxidative responses). - Data indicated acupuncture can influence these pathways across animal models and human studies; the review collated mechanistic findings without providing unified dosing or protocol standards. - Investigators noted changes in pathway activity consistent with improved lipid/glucose handling, reduced inflammatory marker signaling, and modulation of neuroendocrine regulators of appetite. - The review referenced both animal and human research (MeSH terms include Animals and Humans) but did not report specific sample sizes, standardized acupuncture regimens, or uniformly measured clinical endpoints in the abstract. - Authors concluded that acupuncture is a multi-target intervention affecting metabolic and inflammatory networks relevant to obesity, and they argued this provides a theoretical basis for clinical application and individualized weight-loss strategies. - The article is published in Zhen Ci Yan Jiu (2026) and is reported in Chinese; conflicts of interest were declared absent by the authors. - The review cited a broad literature (69 references shown) including guideline and epidemiologic sources, but explicit methodological heterogeneity and gaps in translational evidence were acknowledged implicitly in the abstract.

The pathogenesis involves dysregulated glucose and lipid metabolism, insulin and leptin resistance, chronic low-grade inflammation, oxidative stress, and abnormal fat accumulation. These alterations are closely associated with aberrant regulation of key signaling pathways such as adenosine monophosphate-activated protein kinase (AMPK), Janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3), Toll-like receptor 4/nuclear factor κB (TLR4/NF-κB), and silent information regulator 1 (SIRT1) signaling network. As a multi-target intervention method, acupuncture therapy has made significant progress in recent years in improving obesity-related pathological conditions and signaling pathway imbalances. This article reviews the regulatory effects of acupuncture on signaling pathways such as AMPK, JAK2/STAT3, TLR4/NF-κB, and SIRT1 signaling network, explores its molecular mechanism of action in improving obesity, and provides a scientific theoretical basis for the clinical application of acupuncture therapy and the optimization of individual weight-loss strategies. . **Keywords:** Acupuncture; Molecular mechanism; Obesity; Research progress; Signaling pathway.
