## Executive GIST (TL;DR)
- Rapid detection of **Salmonella** in poultry supply chains was developed to overcome delays in culture-based methods; the study reports a same-day, culture-independent workflow. - The assay workflow combines **immunomagnetic separation (IMS)** to capture cells, **whole-genome amplification (WGA)** to increase nucleic acid yield, and **loop-mediated isothermal amplification (LAMP)** targeting the conserved **invA** locus for detection. - Two readouts were used: fluorescence-based real-time LAMP and phenol red–based colorimetric LAMP; both were integrated and benchmarked against existing workflows. - Analytical sensitivity: the IMS-WGA-real-time LAMP detected 10 CFU g-1 of Salmonella enterica serovar Enteritidis in spiked chicken carcass rinsate with a 90% detection rate (9/10) and in spiked feed with an 80% detection rate (8/10). - Inclusivity and exclusivity: testing used 27 Salmonella enterica strains representing 23 serotypes and six non-Salmonella strains; results showed 100% agreement with invA targeting. - The end-to-end process, from sample preparation to result, required approximately 5 hours and minimal equipment, making it suitable for on-site screening in poultry-related matrices. - Authors: Hyungsuk Oh, Hyunsook Kim, Kun-Ho Seo; affiliations include the Center for One Health, Konkuk University, and Department of Food & Nutrition, Hanyang University, South Korea. - The work emphasizes a rapid, sensitive alternative to culture-based surveillance for poultry carcass rinsate and feed, with potential utility for timely risk management in supply chains. - The authors declared no competing financial interests or personal relationships in the conflict of interest statement.

Therefore, we developed a culture-independent workflow combining immunomagnetic separation (IMS), whole-genome amplification (WGA), and loop-mediated isothermal amplification (LAMP) targeting the conserved invA locus to enable same-day detection of Salmonella in chicken carcass rinsate and poultry feed. Fluorescence-based real-time LAMP and phenol red-based colorimetric LAMP were integrated and benchmarked against existing workflows. The IMS-WGA-real-time LAMP detected 10 CFU g-1 of Salmonella enterica serovar Enteritidis (S. enteritidis) in spiked carcass rinsate with a 90% detection rate (9/10) and in spiked feed with an 80% detection rate (8/10). Inclusivity and exclusivity testing using 27 Salmonella enterica strains with 23 serotypes and six non-Salmonella strains showed 100% agreement with invA targeting. The entire process, from sample preparation to obtaining results, took approximately 5 h and required minimal equipment. Overall, IMS-WGA-LAMP offers a rapid and sensitive alternative for on-site Salmonella screening in poultry-related matrices. **Keywords:** Immunomagnetic separation; Loop-mediated isothermal amplification; Poultry; Salmonella; Whole-genome amplification.
