## Executive GIST (TL;DR)
- The study presents a **multimodal** workflow combining on-axis, high-speed brightfield microscopy with scanning small-angle X-ray scattering (**SAXS**) to monitor irradiation effects on cells during scans. - Investigators integrated the microscope into the SAXS setup at beamline ID13 of ESRF and recorded brightfield images in synchrony with SAXS data acquisition, enabling time-resolved imaging during exposure. - High-brilliance synchrotron sources, nano-focused beams and fast scanning modes allow rapid structural interrogation of hydrated cells, but the resulting high photon flux increases dose and produces observable intracellular damage. - The team observed and quantitatively tracked changes in cellular morphology in situ, using the brightfield channel as a complementary readout to scattering-derived structural information. - Authors noted that this combined approach yields multifaceted information about radiation-induced alterations in biological samples, extending beyond what SAXS alone provides. - The preprint declares no competing interests and lists funding from the German Federal Ministry of Research, the German Research Foundation (DFG), and Germany’s Excellence Strategy (MBExC); specific grant numbers and project IDs are reported. - The source did not report specific irradiation doses, quantitative damage thresholds, exact image-analysis metrics, or stepwise mitigation strategies; those methodological details were not provided in the preprint. - This method targets cellular samples and other biological specimen types exposed to high photon flux during nano-focused, high-brilliance SAXS experiments, with potential for wider adoption in beamline workflows.

Among these techniques, scanning small-angle X-ray scattering (SAXS) is a powerful method for investigating ordered structures within cells. With the recent advent of high-brilliance synchrotron sources, nano-focused X-rays and fast scanning modes, scanning SAXS can now provide structural information from hydrated cells in liquid environments in little time. However, the high photon flux results in an increased dose and pronounced radiation damage to the intracellular structures. Therefore, strategies are required to monitor and quantitatively analyze the extent of damage. Here, we present a multimodal approach that employs an on-axis, high-speed, brightfield microscope to monitor cellular damage during scanning. This microscope is integrated into the scanning SAXS setup at ID13 of ESRF and records images in synchrony with SAXS data acquisition. We are able to observe and quantitatively track changes in cellular morphology \textit{in situ} and thereby obtain complementary information to that provided by scanning SAXS measurements. Our approach opens up the potential for a multifaceted investigation of radiation damage to biological cells and other sample types. ### Competing Interest Statement The authors have declared no competing interest. ## Funder Information Declared German Federal Ministry of Research, Technology and Space (BMFTR), 05K22MG3, 05K25MGA German Research Foundation (DFG), project-ID 449750155 - RTG 2756, projects A2, A7 Germany's Excellence Strategy - EXC 2067, `Multiscale Bioimaging: from Molecular Machines to Networks of Excitable Cells' (MBExC), EXC 2067/1-390729940 DAPHNE4NFDI, 460248799 Copyright The copyright holder for this preprint is the author/funder, who has granted bioRxiv a license to display the preprint in perpetuity.
